O jogo atualmente consiste de todos os 78 caracteres. Sete caráteres, Dark Samus, Margarida, Lucina, Chrom, poço escuro, Richter, e Ken não são dados seus próprios números entre o roster devido a ser lutadores do eco; em vez disso, eles têm o número do lutador que são baseados fora de junto com um símbolo Épsilon (ε) ou apóstrofo ('). O instrutor de Pokémon conta como três personagens, mas só ocupa um local de lista, com a seleção do Pokémon inicial chegando mais tarde. Além disso, este jogo terá 6 personagens DLC, o primeiro personagem DLC sendo Piranha Plant, eo segundo personagem DLC sendo Joker, que é o protagonista de persona 5.

Cada lutador dos quatro jogos anteriores retornou para esta parcela. Por sua vez, a equipe de desenvolvimento afirmou que não haveria muitos recém-chegados. Há onze recém-chegados, com cinco deles sendo Echo Fighters.

A lista inicial consiste no original 8 personagens de Super Smash Bros. com o resto dos personagens sendo unlockable.
